Derivation of Reaction Rate Expression with Substrate Inhibition

Biochemical Engineering

Problem Statement: Consider the following enzymatic reaction mechanism where an enzyme can combine with a substrate molecule to form an active complex, which can further combine with a second substrate molecule to form an inactive complex.

             k1          k5
    E + S <------> ES ------> E + P
             k2

             k3
    ES + S <------> ESS
             k4
List the equations that need to be solved to derive the rate expression with a) the equilibrium assumption and b) the quasi-steady state assumption. Sketch the dependence of reaction rate on substrate concentration.
